Tafsafe with active ingredient tenofovir alafenamide is a newly developed tenofovir prodrug to improve renal safety when compared to tenofovir disoproxil. Both of these prodrugs were first engineered to coat the polar phosphoric acid group on tenofovir using a novel carbonyl oxygen ethyl oxygen binder to improve oral bioavailability and intestinal diffusion. Tenofovir alafenamide is an alanine ester that has been characterized to exhibit low systemic but high intracellular concentrations

Renal tolerability of tenofovir alafenamide was better when compared with its counterpart tenofovir disoproxil. In several clinical trials, tenofovir alafenamide was shown to have 5 times more potent antiviral activity against HIV-1 than tenofovir disoproxil.
